Product Introduction

The Urease Biochemical Tube (for Legionella) is a ready-to-use, pre-sterilized liquid culture medium designed for the biochemical identification of Legionella species, including Legionella pneumophila. Eliminating the need for media preparation, pH adjustment, or sterilization in the laboratory, these glass ampoule-packaged tubes deliver consistent, visual results to support routine Legionella detection in clinical, environmental, and industrial settings.

Test Principle

The assay is based on the ability of bacteria to produce the enzyme urease: Urease-positive bacteria hydrolyze urea into ammonia, raising the medium’s pH and turning the phenol red indicator rose-red (positive). Legionella species do not produce urease, so the medium remains yellow or unchanged (negative). This clear color differentiation allows for straightforward identification and differentiation of Legionella from urease-positive organisms such as Proteus mirabilis.

Procedure and Result Interpretation

Inoculation: Using a sterile loop, transfer a pure culture of the test bacterium into the urease biochemical tube.
Incubation: Incubate aerobically at 36±1°C for 24–120 hours.
Observation: Check the color change of the medium daily.
Interpretation:
  • Positive (+): Medium turns rose-red (indicates urease production).
  • Negative (-): Medium remains yellow or unchanged (no urease production, consistent with Legionella).
  • Blank Control: Uninoculated medium should remain light amber/yellow throughout the incubation period.
